Elastin is a polymeric protein and is the main component of the extracellular matrix of arteries. It is synthesized and secreted as a soluble, single-chain protein (tropoelastin), which undergoes a number of post-translational modifications prior to its organization into an elastic fiber in the extracellular space. Elastin performs a regulatory function during arterial development by controlling proliferation of smooth muscle and stabilizing arterial structure. It imparts elasticity to the connective tissue. During aging, the elasticity of connective tissue is reduced because of the cross-linking of collagenous fibers with elastin. Calpain 1 is an intracellular calcium-dependent protease that cleaves cytoskeletal and sub-membranous proteins. Calpains are non-lysosomal, calcium-activated intracellular cysteine proteases. Calpains mediate specific Ca2 - dependent processes including cell fusion, mitosis and meiosis. Calpains are heterodimers of a small regulatory subunit and one of three large catalytic subunits, designated Calpain 1, Calpain 2 and Calpain p94. Calpastatin regulates Calpain by inhibiting both the proteolytic activity of Calpain and its binding to membranes. Calpastatin exists in two types, tissue type and erythrocyte type, resulting from both alternative splicing and proteolytic processing. Calpain 1 co-localizes with human leukocyte antigen-DR (HLA-DR) on activated microglia in the aging brain. Recognizes protein of 192 kDa, which is identified as Clathrin Heavy Chain. Clathrin is composed of three heavy chains and three light chains, which associate non-covalently to form a triskelion structure. Clathrin heavy chain (HC) is composed of a terminal globular domain, a distal segment and a proximal segment containing a light chain-binding site. The proximal segment of the Clathrin HC protein is essential for interactions between Clathrin heavy chains and light chains, which result in the formation of the triskelion structure. This antibody recognizes a protein of about 37 kDa, which is identified as Erythropoietin (EPO). Erythropoietin is a secreted, glycosylated cytokine hormone composed of four alpha helical bundles. It is the primary factor responsible for regulating erythropoiesis during steady-state conditions and in response to blood loss and hemorrhage in the adult organism. Erythropoietin is synthesized by the kidney and stimulates the proliferation and maturation of bone marrow erythroid precursor cells.
